Effect of Demineralized Bone Matrix Powder from the Femoral Shaft of the Rabbit on the Mouse Myoblast (C2C12)

Abstract
Purpose: The purpose of this study is to verify the effect of demineralized bone matrix powder on the mouse myoblast (C2C12).

Materials and Methods: Bone powder with particles of 500~710 ¥ìm in size has been manufactured by pulverizing the femur of the New Zealand white rabbit using of bone mill and demineralization under 0.5 N HCl solution. The mouse myoblast was cultured with DMEM containing 10% FBS, reaching the confluency of 75%, and then each of demineralized bone matrix (DBM) powder, heated-DBM powder(negative control), or undemineralized bone matrix powder(positive control), respectively. Finally, the test sample was added for further incubation to measure the level of alkaline phosphatase by spectrophotometry at 405 nm.

Results: DBM powder with particles of 500~710 ¥ìm showed 2% concentration of calcium level at 203 minutes. The peak expression level of alkaline phosphatase has reached at 5 days of culture with subsequent decrease of the level.

Conclusion: DBM powder demonstrated potential capacity of bone formation, showing the peak expression of alkaline phosphatase at 5 days of culture with the mouse myoblast.
